Subcutaneous self-administration of oxyntomodulin (400 nmol, three times daily, 30 minutes before meals) for 4 weeks significantly reduces body weight in overweight and obese subjects by reducing energy intake without altering subjective food palatability.
For overweight or obese individuals, a 4-week trial of subcutaneous oxyntomodulin (400 nmol, 3x daily before meals) can lead to significant weight loss (~2.3 kg) by reducing food intake without making food taste worse. While injections are required and nausea is a potential side effect (manageable by dose adjustment), compliance was high in this study. This suggests that mimicking natural satiety hormones is a viable, specific anti-obesity strategy compared to broader neurotransmitter drugs.
Self-administering oxyntomodulin subcutaneously effectively reduced body weight by 2.4% over a 4-week period in healthy overweight and obese volunteers... A significant reduction in energy intake with preservation of subjective food palatability was also observed.
Why this rating
Double-blind, randomized, controlled trial with statistically significant results (P < 0.01), though limited by small sample size (n=26) and short duration (4 weeks).

- Initial treatment for type 2 diabetes should be a combination of metformin and either an SGLT-2 inhibitor or a GLP-1 receptor agonist to achieve cardiorenal protection, rather than monotherapy or older agents like sulfonylureas.Strong
- For patients with specific monogenic obesity syndromes (leptin deficiency, POMC/PCSK1/LEPR mutations), targeted pharmacotherapy (recombinant leptin or setmelanotide) is highly effective and should be prioritized, unlike in polygenic obesity.Strong
- Continued weekly administration of 2.4 mg subcutaneous semaglutide prevents weight regain and promotes further weight loss in adults with overweight or obesity, whereas switching to placebo results in significant weight regain.Strong
