Daily oral administration of MK-677 (10-25 mg) to healthy elderly subjects (64-81 years) significantly increases 24-hour mean GH concentrations and serum IGF-I levels in a dose-dependent manner by enhancing the amplitude of pulsatile GH secretion without changing pulse frequency.

For healthy older adults, taking 10-25 mg of oral MK-677 daily for 2-4 weeks significantly boosts Growth Hormone and IGF-1 levels, bringing them closer to young adult ranges. This is achieved by increasing the size of natural GH pulses rather than creating constant high levels. While it may slightly raise fasting blood glucose, it was generally well-tolerated in this short-term study, offering a non-injectable alternative to GH therapy.
