To date, MR studies using BMI-associated variants have provided evidence of a causal effect of greater adiposity on a number of indicators of reduced cardiovascular health, including increased blood pressure (43), increased fasting glucose and insulin (44), decreased HDL-C (44) and increased systemic inflammation (45). Causal inference with respect to complex diseases is challenging however, a causal role for increased adiposity has been evidenced for type 2 diabetes T2D) (44) and ischaemic heart disease (IHD) (46).
