Metformin, a common diabetes drug, may delay aging and limit age-associated diseases by inducing metabolism associated with dietary restriction and blocking NF-κB activity.
Metformin is being studied for its potential to extend healthspan by mimicking the metabolic effects of calorie restriction. A major FDA-supported clinical trial is currently investigating its use in non-diabetic individuals for anti-aging purposes.
ModerateConditionalMEDIUM confidence
A potential mediator of metformin benefits is the AMP-activated kinase (AMPK) and metformin can be viewed as DR-like compound [76].
Why this rating
Validated in rodents and C. elegans; human clinical trial 'Metformin, Anti-aging' is mentioned as a landmark event but results are not detailed.

- Calorie restriction (CR) attenuates the age-related upregulation of proinflammatory cytokines (NF-κB, IL-β, IL-6, and TNF-α), serving as a primary intervention to counteract inflamm-aging.Moderate
- Resveratrol acts as a SIRT1 activator to inhibit NF-κB activation and suppress proinflammatory molecules (IL-1β, IL-6) in a dose-dependent manner, thereby inhibiting inflamm-aging.Moderate
